產品概述
粒細胞集落刺激因子(G-CSF)是在嗜中性粒性白細胞系的造血細胞的增殖、分化和活化起重要作用的多向性細胞因子。 G-CSF是通過活化的單核細胞,巨噬細胞,成纖維細胞,內皮細胞,星型膠質細胞,成骨細胞,骨髓基質細胞等細胞產生的。除此之外,各種癌細胞系和成髓細胞白血病細胞也能組成性的產生 G-CSF。已經表明G-CSF和G-CSF受體能夠通過胎盤,蛻膜和子宮內膜腺體細胞在懷孕期間產生,這表明G-CSF有可能在胎盤和蛻膜中起重要作用。
在人類中,兩個G-CSF的cDNA編碼207和204個氨基酸殘基的前體蛋白質已經得到了分離。這兩個蛋白質,除了在成熟蛋白質的N端起的35個氨基酸以后的三個氨基酸差異外, 都有一段30個氨基酸的信號肽并且有一致的氨基酸序列。
產品原理
本實驗采用雙抗體夾心ELISA法。抗人G-CSF單抗包被于酶標板上, 標本和標準品中的G-CSF會與單抗結合,游離的成分被洗去;加入生物素化的抗人G-CSF抗體,生物素化抗人G-CSF抗體與包被在酶標板上的抗人G-CSF單抗結合而形成免疫復合物,游離的成分被洗去;加入辣根過氧化物酶標記的親合素,親合素與生物素特異性結合,游離的成分被洗去。加入顯色底物(顯色劑),若反應孔中有G-CSF,辣根過氧化物酶會使無色的顯色劑變藍,加終止液變黃。在450nm處測OD值, G-CSF濃度與OD450值之間呈正相關,可通過繪制標準曲線求出標本中G-CSF濃度。
